Wever got her start as a performer in indie, low-budget films. She has appeared in theatrical performances, including two off-Broadway plays alongside Deidre O’Connell: Cavedweller and Smashing by Brooke Berman. Since the start of her acting career, Wever has appeared in a large number of major motion pictures, including Into the Wild, Neal Cassady, Michael Clayton, Series 7: The Contenders, Signs, The Adventures of Sebastian Cole, Bringing Rain, Tiny Furniture, and All I Wanna Do, among others.

She has appeared in several TV shows as a guest star, including The Wire, The Good Wife, Conviction, Law & Order: Criminal Intent, and New Girl. Alongside Rachel Blanchard, Austin Nichols, and Shiri Appleby, she co-starred in Ed Zwick’s misguided pilot for the television series Quarterlife. Additionally, she played Suzanne in a recurrent episode of Studio 60 on the Sunset Strip.

Wever is most well-known for her part as Zoey Barkow in the Showtime television series Nurse Jackie, which debuted in June 2009. According to the official Nurse Jackie website, Zoey is “a comic contrast to Edie Falco’s hard-bitten (and prescription drug-addicted) titular heroine.” She is defined as “an irrepressibly perky trauma nurse.”  Wever’s performance in the show was praised highly by the critics.

Wever was nominated for a Primetime Emmy Award in 2012 and 2013 for her performance in Nurse Jackie. In 2013, she won the award and gave a noteworthy acceptance speech that was limited to her expression of shock: “Thanks a lot. I greatly appreciate it. I must leave. Bye.”

Denise Cloyd, played by Wever, appeared on The Walking Dead in 2016. Her character’s demise near the end of the show’s sixth season was contentious and infuriated many fans, particularly members of the LGBT community, on social media.

Wever portrayed Mary Agnes McNue, a severe mayor of a village where women predominate, in the 2017 miniseries Godless. She admitted to being terrified by the part in interviews: “I spent so much of this shoot thinking I would come out like a fool, that nobody would buy me as this and it wouldn’t be real.”

She received an Emmy for the part in 2018. Wever and Domhnall Gleeson co-star in the HBO black comedy thriller series Run beginning in 2020. It was discontinued following one season.
